I just saw this thread. So, evidently there are at least four Yamnaya who are R1b-L51. That is HUGE. Some of us, including me, have been waiting for this kind of news for many years.

I am hesitant to believe it right now, because it just seems too good to be true, but if it is true, THANK GOD!

The paper, if I understand correctly, claims that it was the Progress/Vonyuchka type people (the CLV cline) who spoke Basal PIE (i.e. PIE before the Anatolian branch split and headed for central Anatolia), making the N Caucasus/Lower Volga region, NOT Sredny Stog/Dnieper-Don, the overall PIE urheimat. I'll have to read the paper first, though, heh. I wonder if they explained that Central Asian ancestry in IE speaking nomads.

Yes, but... Progress was not homogeneous autosomally.

Regarding Progress 2:

This ancestry profile is known as 'Eneolithic Steppe' ancestry, or 'pre-Yamnaya ancestry', and is represented by ancient individuals from the Khvalynsk II and Progress 2 archaeological sites. These individuals are chronologically intermediate between EHGs and the later Yamnaya population, and harbour very variable proportions of CHG ancestry.[22][23][10]

This early, 'pre-Yamnaya' ancestry was first detected in Eneolithic individuals at the Khvalynsk II cemetery and directly north of the Caucasus mountains at the Progress 2 archaeological site; this ancestry is also detected in individuals of the Steppe Maykop culture, but with additional Siberian and Native American-related admixture.[25][5]

Three individuals from the Progress 2 site in the foothills north of the Caucasus also harbour EHG and CHG related ancestry, and are genetically similar to Eneolithic individuals from Khvalynsk II but with higher levels of CHG-related ancestry that are comparable to the later Yamnaya population.[5][26]

Archaeologist David Anthony speculates that the Khvalynsk/Progress-2 mating network, located between the middle Volga and the North Caucasus foothills, makes a "plausible genetic ancestor for Yamnaya".[26]

No, Caucasus ancestry is even smaller in BA Anatolia than the steppe ancestry. ~88 % is the Mesopotamian ancestry and most of the rest is the steppe ancestry. This is explained thoroughly in the end of their supplementary PDF.

Do we know anything more about these Mesopotamian ancestry? As far as I can tell no unipaternals, but what about autosomal?

I2a from the Dneper spoke likely spoke a euro hunter language tilted toward the WHG

I just wonder how good they are to differentiate ANF ancestry on both sides of the sea, especially in the Anatolian context, which got such relatively minor admixture from the steppe. That means any WHG-EEF would be pretty minor as well, regardless, unless they assume all steppe groups which came from the West must have had very high levels of EEF-WHG, for which there is, in my opinion, no reason. We already see that there all kind of proportions of steppe : WHG : EEF around.

The best support possible is therefore always following the uniparentals, because they are unambiguous if such evidence is present and not lost due to drift or other factors.

I wonder what the climate in the Northeast Caucasus/Lower Volga region was like during the PIE era.

At present, the region is mostly a desert or at least semi-arid, and much of the population consists of speakers of Mongolic (Kalmyk) or Turkic languages (e.g. Kumyk, Nogai) whose indigeneity is highly questionable.

Is that really the sort of region from which one should expect a major language family to have expanded?

Of course, the real evidence, such as Y-DNA, mtDNA, and autosomal DNA, from ancient human specimens is most important to determining what has actually occurred there. I just find it to be a rather unlikely region of origin a priori.

That's one hell of a lot of R1b. The I2-L699 is kind of fascinating in Yamnaya too given that it made it into western Europe, yet has been absent from Beaker to this point. 
Out of the three theories, I understand how the western route is rejected based on the drop in EEF and Balkan, but the eastern steppe route with V1636 being a Proto-Anatolian marker makes the most sense. The uniparentals in the steppe don't really support any other scenario, and how or why is it that the dozens of language isolates in the Caucasus mountains didn't also have such a profound effect on the steppe, assuming that's the origin of early PIE? Seems like special pleading, where as the opposite doesn't require much at all given that ancestry can easily change over a handful of generations with locals, but language change isn't nearly as straight forward.

Given the limited steppe in Anatolia is it not safe to say that wave 1 that brought Indo-Anatolian to Anatolia was primarily CHG?

No, Caucasus ancestry is even smaller in BA Anatolia than the steppe ancestry. ~88 % is the Mesopotamian ancestry and most of the rest is the steppe ancestry. This is explained thoroughly in the end of their supplementary PDF.

They support the Caucasus route for the Anatolian branch because their steppe ancestry represents the Caucasus-Lower Volga cline (CLV) and because the Anatolians lack the European ancestries WHG and the admixed EEF. 

So, it seems (roughly simplified) that the Indo-Anatolian admixture of Caucasus and EHG ancestries spread to the south across the Caucasus, where they admixed with the Mesopotamian ancestry, which became prevailing, and this population spread west to Anatolia, were earlier Anatolian Farmer ancestry was prevailing. 

Because there is no conclusive linguistic evidence supporting either route around the Black Sea, the genetic results weigh the situation to the Caucasus route.

But if the Authors are arguing for a first wave spread of BPgroup ancestry being responsible for Indo Anatolian language spread in Anatolia, what does the BA ratio of CHG-EHG have to do with that first wave? 

CLV cline people with Lower Volga ancestry contributed four fifths of the ancestry of the Yamnaya, but also, entering Anatolia from the east, contributed at least a tenth of the ancestry of Bronze Age Central Anatolians, where the Hittite language, related to the Indo-European languages spread by the Yamnaya, was spoken. We thus propose that the final unity of the speakers of the “Proto-Indo-Anatolian” ancestral language of both Anatolian and Indo-European languages can be traced to CLV cline people sometime between 4400-4000 BCE.

That means any WHG-EEF would be pretty minor as well, regardless, unless they assume all steppe groups which came from the West must have had very high levels of EEF-WHG, for which there is, in my opinion, no reason. But they see clearly 3 % of the Caucasus ancestry in their modeling, so probably they would see equally small proportions of the Farmer ancestry.

Yes, continuity would be undeniable with uniparental lineages. But new lineages spread and old lineages get replaced and disappear. In every step of language expansion the genetic composition of the language carriers can be different than in the previous step. This concerns both autosomal ancestries and uniparental lineages. It is normal for old, widespread language families to have no single ancestry component or uniparental lineage which would be present in all the populations. But the growing resolution of the ancient DNA samples might help to fill the gaps.

While I've supported for a long time the eastern steppe route idea for Anatolian, we have no evidence of whether PIE is EHG or CHG, and never will. The buck stops here unfortunately. All we can do is speculate.
